There are seven ties involving teams from the Southern League, with the game between two in-form sides in Premier Division South rivals Metropolitan Police and Farnborough being one of the stand-outs.
Unbeaten Division One South side Sholing will also fancy causing an upset with they visit a Premier Division South Tiverton Town outfit who have won only once in the league to date.
A couple of teams face lower league opposition, with Division One Central Aylesbury United welcoming Uhlsport Hellenic League Premier Division side Bishops Cleeve to the Meadow.
Cleeve are former members of the Southern Legaue, being promoted in 2006 and spending twelve seasons in either the Midlands or South West sections before being relegated back to the Hellenic League at the end of the 2017/18 campaign.
Division One South club Cirencester Town, who have yet to lose in 2021/22 to date but have drawn their last two, host Sydenhams Football League (Wessex) Premier side Hamworthy United at the Corinium Stadium.
The Hammers have already played four FA Cup ties this season, beating rivals Brockenhurst in the Extra Preliminary Round, Westbury United in the Preliminary Round and then causing an upset in the First Qualifying Round by knocking out Premier Division South side Merthyr Town after a replay, so will not be taken lightly by the Centurians.
Vanarama National League North and South teams are involved at this stage of the competition and several Southern League face tough tests, including Premier Division Central Coalville Town, who travel to St James`s Park to take on Brackley Town, who are currently second in the north section.
Division One South side Frome Town will quietly fancy their chances of causing a surprise when they host National League South outfit Oxford City at Badger`s Hill.
City have only lost once in the league so far though, so it will be a tough ask for the Somerset side.
Premier Division South side Harrow Borough started the season indifferently but have now won four on the spin in league and cup.
They travel the relatively short distance to Park View Road to take on National League South outfit Welling United, who have won just the once in league action so far.
There is sure to be a decent crowd at Wodson Park where Division One Central newcomers Ware welcome their Hertfordshire neighbours from the National League South, Hemel Hempstead Town.
Ware have settled well since being switched from the Isthmian League South Central and enjoyed a fine 3-2 win at The Eyrie against fancied Bedford Town on Tuesday night.
And if there`s going to be an upset to predict, this could be it as the Tudors have lost five of their six league matches to date, only picking up their first point of 2021/22 last weekend at home to Eastbourne Borough in a 2-2 draw.
Premier Division South side Poole Town will also fancy their chances of being in the draw on Monday when they welcome Hemel`s league rivals Chippenham Town to the Black Gold Stadium.
The Bluebirds, promoted from the Southern League after being Premier Division champions in 2016/17, have won twice in the league to date, but lost at home to Chelmsford City last Saturday.
Poole`s league rivals Walton Casuals also have National League South visitors on Saturday as Hampton & Richmond Borough head to the Elmbridge Sports Hub.
It`s tough to know what to predict here as the Beavers have won two, drawn two and lost two of their league games to date!
One where I would stick my neck out and predict an `upset` is hopefully at the SKYex Community Stadium where Hayes & Yeading United, who possess a 100% record to date, welcome National League South side Tonbridge Angels.
Angels, who were members of the Southern League for many years, have won just the once this season at the end of August.
Hayes` league rivals Beaconsfield Town face a tough test, however, as they make the trip down to Hampshire to play National League South outfit Havant & Waterlooville, buoyed though by the fact that the Hawks lost at home last Saturday!
Another Premier Division South club, Truro City, make the journey to Bulpit Lane to play another National League South side, Hungerford Town, who have only played four games so far this term, winning two and losing two.
Lymington Town, only promoted to Division One South in the summer from the Wessex League, will be fancying their chances of causing an upset against a club whose former guise was built around causing major cup shocks themselves, National League North Hereford FC.
The current Bulls, formed in 2015 to replace the folded United version, came through the Southern League on its way to their rapid rise to Step 2 of the non-League Pyramid just three years later.
The capacity at the Sports Ground could well be severely tested as Hereford are always well supported, despite being currently bottom of the table without a win to date.
